INDICATION

Probeta-N Drop is a combination ophthalmic solution containing a corticosteroid and an aminoglycoside antibiotic. It is indicated for steroid-responsive inflammatory conditions of the eye where bacterial infection or risk of infection exists. It is prescribed for bacterial conjunctivitis with inflammation, blepharoconjunctivitis, keratitis, iridocyclitis, and post-operative inflammation following ocular surgery.

MECHANISM OF ACTION

Betamethasone is a potent synthetic corticosteroid that suppresses ocular inflammation by inhibiting phospholipase A2, reducing prostaglandin and leukotriene synthesis, stabilizing cell membranes, and decreasing vascular permeability. Neomycin is an aminoglycoside antibiotic that inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit. The combination provides concurrent anti-inflammatory and antibacterial action.

DOSAGE AND ADMINISTRATION

     Instill 1 to 2 drops in the affected eye(s) every 4 to 6 hours

     For severe inflammation: instill every 1 to 2 hours initially, reducing frequency as the condition improves

     Do not exceed 14 days of treatment without ophthalmologist review

     Shake well if suspension

     Wash hands before and after application

SIDE EFFECTS

Common:

     Mild burning or stinging upon instillation

     Temporary blurred vision

     Eye irritation

Less common:

     Increased intraocular pressure with prolonged corticosteroid use

     Posterior subcapsular cataract with extended therapy

     Secondary infection due to immunosuppression

     Neomycin sensitization causing allergic contact dermatitis

NOT RECOMMENDED IN

     Viral eye infections including herpes simplex keratitis, vaccinia, and varicella

     Fungal or mycobacterial eye infections

     Known hypersensitivity to betamethasone, neomycin, or any aminoglycoside

     Patients with a history of neomycin allergy

DRUG INTERACTIONS

Concurrent use with other corticosteroid eye drops may increase the risk of elevated IOP. Avoid concurrent topical aminoglycosides. Remove contact lenses before application.

STORAGE INSTRUCTIONS

Store below 25 degrees Celsius. Protect from light. Do not freeze. Discard 4 weeks after opening. Keep out of reach of children.
