INDICATION

MECHANISM OF ACTION
Omeprazole is a substituted benzimidazole that irreversibly inhibits the hydrogen-potassium ATPase (proton pump) on the parietal cell surface. The suspension provides the same pharmacological action as capsule formulations but in a format suitable for patients who cannot swallow solid dosage forms.
DOSAGE AND ADMINISTRATION
● Infants 1 to 11 months (3 to 5kg): 2.5mg once daily
● Infants 1 to 11 months (5 to 10kg): 5mg once daily
● Children 1 to 16 years (10 to 20kg): 10mg once daily
● Children 1 to 16 years (over 20kg): 20mg once daily
● Adults: 20 to 40mg once daily, depending on the condition
● Administer on an empty stomach, 30 minutes before a meal; shake well before each dose
SIDE EFFECTS
Common:
● Headache
● Diarrhea or constipation
● Nausea and abdominal pain
● Flatulence
Less common:
● Hypomagnesemia with prolonged use
● Clostridium difficile-associated diarrhea
● Increased risk of bone fractures with long-term use
● Vitamin B12 and iron deficiency with chronic therapy
● Acute interstitial nephritis (rare)
NOT RECOMMENDED IN
● Known hypersensitivity to omeprazole or any substituted benzimidazole
● Concurrent use with nelfinavir or rilpivirine
● Patients with osteoporosis on long-term PPI therapy without calcium and vitamin D supplementation
DRUG INTERACTIONS
Omeprazole reduces gastric acidity and may decrease absorption of drugs requiring acid pH, including ketoconazole, itraconazole, and iron salts. Omeprazole inhibits CYP2C19 and may increase levels of diazepam, phenytoin, and cilostazol. Clopidogrel antiplatelet effect may be reduced. Concurrent use with methotrexate may increase methotrexate toxicity.
